That would be the vaccuum inlet for your petcock vaccuum line.
Plug it if you bypassed the membrane or petcock all together, or run a new line from that inlet to your petcock. If you dont your bike will run like ass, or you won't get fuel.

I'm sorry for the questions just new to bikes n working on anything. The membrane is the 2 top things u have to cover with the plates

@Asencion Ortega Nope, the membrane would have been inside your petcock. Looks like you have it bypassed completely with that ball valve so plugging that vaccuum inlet should be all you need to worry about.
